ES Notes Special Edition July 2011

After sucessfully exhibiting at the SFC 2011 and attending the day dedicated to SFC purification at the PREP 2011 meeting and seeing the increased interest and anticipation we decided to bring you a dedicated SFC e-Newsletter - aimed at bringing you technical information and new product introductions. You are one of an exclusive group of scientists selected to recieve this introduction issue, with features on:

  • GreenSep™ Basic - GreenSep Basic is the SFC column ideally suited for the retention and rapid separation of chemicals containing amine groups. GreenSep Basic can easily replace conventional stationary phases used in SFC and deliver superior performance.

  • Achiral SFC - joint poster presented at SFC 2011 entitled:

A Systematic Study of Achiral SFC Stationary Phases Using a Computational Calculated Molecular Diversity Model

Matt Przybyciel, ES Industries, West Berlin, NJ and

Ray McClain, Dept of Medicinal Chemistry, Merck Research Laboratories

  • GreenSep GCS Guard Column System - GreenSep GCS is packed with highly deactivated high performance silica particles and is designed to maintain the chromatographic integrity of the system.

All designed to be technically informative and practically useful.

The GreenSep Basic stationary phase based on imidazole chemistry, providing a highly basic character, which proves superior to conventional stationary phases (such as diol, cyano etc...) in the areas of separation selectivity, peak shape and loading capacity. The data shown is a prime example of the superior peak shape performance and separation capacity obtainable with the GreenSep Basic column with SFC. The data also compares GreenSep Basic with GreenSep Ethyl Pyridine and demonstrates the superior retention capability that GreenSep Basic can deliver to the SFC chromatographer for chemicals that are functionalized with amine groups and acidic groups. GreenSep Basic offers you, the chromatographer, greater flexibility in developing SFC separations and purifications.

In recent years a number of achiral stationary phases have been developed specifically for achiral SFC. Not all of these columns are well-suited for all structural classes of compounds In order to assess the selectivity of each column, a structurally diverse set of chemicals were selected within four distinct functional group classes (carboxylic acids, amines, alcohols and amides) for evaluation.

Fifteen compounds for each of the four chemical classes were analyzed on eight different achiral stationary phases, in both their end-capped, and non end-capped forms. The resulting data allows us to make recommendations for matching achiral stationary phases to chemical classes. In addition, the data provides an insight into the effect of end-capping in SFC.

GreenSep GCS is a new guard column system developed specifically for use with preparative SFC columns.

The GreenSep GCS employs hardware specifically designed for SFC chromatography and is packaged with a high performance coupler for attachment to the SFC chromatographic column. GreenSep GCS is packed with highly deactivated high performance silica particles and is designed to maintain the chromatographic integrity of the system. A chromatogram using a GreenSep GCS with a GreenSep Pyridyl Amide preparative column is shown. The peaks in the chromatogram are unaffected by the high performance GreenSep GCS guard column.
